Determine variation of specificity across genomic variants

For a given gRNA, does specificity vary across genomic variants of an organism? If so, what's the best way to quantify these differences?

An idea that came up was that we could take published clinical findings as ground truth and train an ML model on it to find a good correlate for specificity. To reduce the number of free parameters, one could look at just the inter-gene of intra-gene confounding effects.
